Maintaining healthy bowel movements is important for overall digestive wellness, as irregularity can contribute to upper gastrointestinal discomfort. In this study, bowel habits were assessed using the Bristol Stool Form Scale. At baseline, participants receiving Asafin (250 mg/day) predominantly reported hard stools, with 65% classified as type 1 and 35% as type 2. By day 8, stool consistency improved substantially, shifting to 26% type 1, 12% type 2, 58% type 3, and 4% type 4. These improvements became even more pronounced by day 14, with only 25% remaining in types 1 and 2, while 75% of participants achieved well-formed stools classified as types 3 and 4. Overall, Asafin supplementation supported normalization of stool form within two weeks, promoting bowel regularity and ease of passage.⁽²⁾

Firmicutes and Bacteroidetes are the two dominant bacterial phyla in the adult gut, together accounting for approximately 90% of the microbiota. These bacteria contribute to intestinal wellness through the production of short-chain fatty acids (SCFAs), including acetate, propionate, and butyrate, which support colonocyte function and help maintain healthy immune and inflammatory balance. The Firmicutes-to-Bacteroidetes (F/B) ratio is commonly used as a broad indicator of microbiome composition. While there is no single ideal F/B ratio, a more balanced ratio is generally associated with greater microbial diversity and a healthier gut environment. In this study, supplementation with Asafin at 250 mg/day for two weeks reduced the F/B ratio by 71.9% (from 4.64 to 1.30), representing a 3.66-fold decrease. This shift moved participants toward a range more commonly observed in balanced adult microbiomes, supporting a healthier, more resilient gut ecosystem.⁽²⁾

The gut–brain axis represents a bidirectional communication network in which gastrointestinal signaling, inflammatory balance, and microbiome composition influence stress resilience and cognitive performance. Supplementation with Asafin at 250 mg once daily for two weeks significantly improved cognitive reaction time. By day 14, reaction speed improved by 45% versus placebo and by 44% versus baseline, decreasing from 722.37 ± 121.42 ms to 399.51 ± 28.48 ms compared with placebo, and from 713.59 ± 111.55 ms to 399.51 ± 28.48 ms from baseline (treatment effect sizes 0.738 and 0.664, respectively; both p < 0.001). No meaningful improvement was observed in the placebo group (p = 0.717). Faster reaction times reflect improved cognitive processing speed, supporting mental agility, focus, and the connection between gut health and cognitive performance.⁽²⁾

In the same 14-day randomized, double-blind, placebo-controlled study, daily supplementation with Asafin at 250 mg significantly supported sleep quality, as measured by the Bergen Sleep Scale (BIS). By day 14, BIS scores decreased by 10.89 ± 3.23 points from baseline (p < 0.001), with a significant difference compared to placebo (ΔBIS = 10.0 ± 2.61, p < 0.001). Asafin supported a 45% improvement from baseline and a 42% improvement compared with placebo. In contrast, the placebo group showed only a modest 6% change. These findings suggest that supporting gastrointestinal comfort and gut–brain balance may promote healthy sleep quality, an important component of overall well-being, mood balance, and daytime cognitive performance.⁽²⁾
